You'd have to be pretty damn fit to ride the loop in a day. If you're taking a couple of days, then you may as well walk it; there's no way you're riding some of those grades with a fully loaded bike.
I've taken 4 days and driven it (and camped) with my kids and thought I'd one day come back: taking a week or so, driving into some of the basins and nabbing a few 14K peaks, hanging out in some of the most spectacular scenery in the US is a worthwhile trip. Biking it is a bit of a waste IMHO.
If I'd resources, I'd take loaded motorcycles on the loop. Scoot quickly from one location to another. In early July, seems like EVERYONE in America with a jeep or SUV or ATV is on that loop! Between the dust, the traffic, the fumes, and the crowds, never mind the sheer effort, biking it would certainly blow big time. Other option: wait till winter and ski it.
